Yes it does. I use one on my S5. You will want to move your needle position to the left.

Q: Will this fit my Janome HD 3000?

It will if your machine is a 9mm. My MC6650 is However most are 7mm

Q: Will it work on Janome JP 720

I have an MC 6550. It connects to the bar so I think it would. If it were just the foot, I have more issues with that.

Q: Will this work on a janome mc9900, and does it require a professional grade needle plate? If so will the needle plate fit the same machine?

I don't know about the mc9900, but I have a Skyline S6 with Acufeed Flex and it works fine when I use my straight stitch needle plate and set the needle to "04" or left position. This foot is for high shank machines only. My machine doesn't work with the professional grade needle plate because it doesn't have the firmware that is activated by that plate (it keeps the needle in the left position and keeps you from using stitches that would break a needle). However, it's no big deal if I forget to set the needle to the left position--the hole on the right side of the foot is exactly where the center position is, so no harm done. I love this foot. It works much better than the single Acufeed Flex foot because the whole foot is over the left set of feed dogs. For some reason, that seems to work better, as some reviewers have noted.
