Do we have to have expansion 1 first in order to add expansion 2?

Depends... If you want to utilize the pink 6th player parts then yes. The Base game has 5 player colors, the first expansion adds a 6th pink color. So if you don't want/care about having a 6th player then you are fine.

Do the Zman expansions work with a Rio Grande original?

i have played both and they look and fit the same.

I have the older version of Carcassonne, the art looks different on the front cover, are tiles different too? or will it match up with the old game?

Same here. The colors are a little bit more vibrant on the newer versions. But it does not affect gameplay.

Will it work with the 20th anniversary edition?

I would think it should since all the other Carcassonne have worked together
