Tank was pumped last week,b leach field is pretty mucked up.Can I drop a tab or two in the distribution box or do I have to wait until tank is filled?

Yes - dropping a tablet or two directly into the distribution box should be very effective in cleaning up your leach field. Two items to check - are there roots that might have grown into your drainpipes to the leach field? If so - use a root killer to remove them. Second item, make sure there is some water at the distribution box for the tablets to dissolve into. If not, pre-dissolve the tablets into a bucket of water, then pour that into the distribution box.

Can I use with root killer too? Not sure if my drainfield issue is from roots or sludge.

Yes, if roots are a potential problem (as is very common) you should use a commercially available root killer. These are available at most local hardware stores. Use the root killer first, then wait 24 to 48 hours and follow with the Doctor Pooper Dran Field Cleaner. If you have already used the DFC, go ahead and apply the root killer, then follow with another treatment of DFC.

Do I have to use this in all three toilets?

It works in the septic system not the toilet.
