Yes, if you wanted to use these bottles for more than just collection and storage you will need the collar and nipple. We have used the medela collar/nipple and also the regular Dr. Browns collar/nipples fit.

how many hours does it take to thaw frozen breastmilk in the refrigerator using these medela bottles?

It typically takes us about 8 hours to defrostin a refrigerator but you can also run these under cold water. Never defrost under hot water because it creates improper heat distribution. You can spoil the milk in direct contact while the core is still frozen.

does it come with the box? any sterilizer bags as the bonus?

Yes, the six bottles are in a box. And in the box there is an extra plastic bag, the bottles can be put into it, then you can sterilize them in the microwave oven. It seems can be used for twenty times.
