Will this work in sand/desert?

No. It’s a kids toy designed for rigid surfaces. Will work on short grass but would not recommend on anything else as the electric motors / tires will not provide required traction or drive for such softer surfaces.

Can the plastic wheels be replaced with rubber wheels?

Hello! Thank you for asking, no I don't believe so, how it comes, is how it is, it is a wonderful quad, and the springs cushion the ring accordingly, we love ours!!!

It's a 12V round hole charger, if you don't know, you can take a look at the replacement I bought asin: b099ph6787, it helped my daughter ride toy very well.
