What's different about these than the other nenesupply duckbill valves?

I found the suction better and pumping session was shorter when compared to duckbill valves. Duckbill valves also have shorter life compared to these valves. First couple of pumping sessions may hurt a little but later on it'll be fine. I used these valves for my Spectra pump

Do these work with medela Lactina?

I'm not sure about that model, but it did not work with my Medela In Style. They fit into the valves but produced no suction, so beware

Will this work with a Spectra pump?

They should, I use them with the NeneSupply and Maymom one piece flanges which I believe are the same size as the Spectra ones.

Will these work with my Spectra 2 pump?

Some people say that they do, but they're not meant to. and I and 2 other women I know pumped significantly less with these on the spectra and the madela. Regular duckbills with significant better And fit medela and spectra
